It Takes 150uL To Fill A NovaSeq S4 Flowcell
In a previous post I tried to estimate how long each sub-step of an Illumina cycle took. I was looking at an S1 flowcell there and kind of just slapped in 15 seconds for washes. This is not a very well informed estimated. I just assume you want to flush a reasonable amount of fluid through the flowcell during washes. Giving the time taken by other steps 15s seemed “reasonable”.
I decided to do some tests with an old NovaSeq 6000 S4 flowcell. Turns out it takes ~150uL and 3 seconds to fill a lane with TE buffer and some food coloring:
