Yesterday I wrote about the PacBio sensor architecture and discussed some thoughts on the evolution of the Sequel, Sequel IIe and Revio. Today I’m going to discuss the other presentation I mentioned briefly and ponder upon PacBio a little.

First I wanted to mentioned a quick follow up to my subread experiments post. I have some more experiments planned here, but I have tried running DeepConsensus on the CCS/read set with pulse information removed. DeepConsensus essentially failed to produce any improvement in accuracy here, suggesting that1 most of what DeepConsensus is doing is incorporating information from the pulse durations/time between pulses.
