Recently a few people have been asking me about PacBio’s platform. Answers to many of these questions would require raw data. Raw data (“Movies”), or even raw subreads do not appear to be available on the Revio.
Some patents provide heat maps of older (Sequel) chips. These are quiet interesting in themselves:
Details of the experiment are in the footnote below1. The heat map is quite interesting in itself. It shows SNR across the chip. And we can see the following features:
Rounded corners: I assume caused by the manifold coupling to the chip (rounded corners are often used to avoid voids in which reagents can diffuse).
Higher SNR around the edges: This seems weird, I assume some kind of reflection of excitation around the manifold?
A rectangular pattern overlayed on the chip. I assume some kind of artifact of the readout system.
This is interesting in itself, but can we generate similar heat maps for Revio data? To some degrees, Yes.
